O fibrinogênio é uma proteína de fase aguda que faz parte da cascata de coagulação de proteínas. O resultado final da cascata é a produção de trombina, que converte fibrinogênio em fibrina. A trombina proteolisa rapidamente o fibrinogênio, liberando o fibrinopeptídeo A. A perda desse pequeno peptídeo não é suficiente para tornar a molécula de fibrina resultante insolúvel, mas tende a formar complexos com as moléculas adjacentes de fibrina e fibrinogênio. Em seguida, a trombina cliva da fibrina um segundo peptídeo, o fibrinopeptídeo B, e, então, os monômeros de fibrina formados se polimerizam espontaneamente para formar um gel insolúvel. A fibrina polimerizada é mantida íntegra por forças não covalentes e eletrostáticas e estabilizada pela enzima de transamidação, fator XIIIa, que é produzida pela ação da trombina sobre o fator XIII. Os agregados insolúveis de fibrina (coágulos) e as plaquetas agregadas bloqueiam, então, o vaso sanguíneo danificado e impedem sangramento adicional. A quantidade de fibrinogênio no plasma pode funcionar como um indicador não específico da presença ou ausência de um processo inflamatório no organismo. O fibrinogênio originário de qualquer mamífero será clivado por uma trombina originária de qualquer mamífero.

Fibrinogen is an acute-phase inflammatory protein, which plays an essential role in blood clotting. Coronary heart disease is linked to the plasma levels of fibrinogen in Chinese population, and might have potential as a marker and risk factor for the same.[1] A -455 allele of fibrinogen β (FGB) chain is linked with poor survival in Caucasian women with Finnish stroke.[2] Ischemic stroke preceded by hyperfibrinogenemia has poor outcome, which is independent of the baseline fibrinogen concentration.[3]

General description
Fibrinogen was the first blood coagulation factor to be identified, and it is a precursor of fibrin protein. This protein is composed of two chains, each of which is composed of three chains- α, β and γ. These three subunits are coded by three genes located in the human chromosomal locus 4q23-q32, and spans 51kb.[1]
